Study 1278 :Get $1,575 - STUDY TO ASSESS EFFECTS OF PF-04457845 ON BOLD FUNCTIONAL MRI IN SUBJECTS WITH PTSD New York

Type of Study | BOLD FUNCTIONAL MRI IN SUBJECTS WITH PTSD | Aim of Study | PF-04457845 is a fatty acid amide hydrolase (FAAH) inhibitor. FAAH is responsible for hydrolyzing the fatty acid amide, anandamide (AEA). Stress induces increases in FAAH activity which result in decreased brain levels of the endocannabinoid (eCB), AEA and reduced signaling via cannabinoid type 1 receptors (CB1). Deficits in eCB signaling may play a key role in the etiology of PTSD and may mediate important components of the PTSD phenotype.
Our hypothesis is that the increase in brain AEA levels achieved by inhibition of FAAH will restore signaling through CB1 in a regionally specific manner and thus alleviate the effects of stress.
| Inclusion Criteria | 1. Evidence of a personally signed and dated informed consent document indicating that the subject has been informed of all pertinent aspects of the study.
2. Subjects who are willing and able to comply with scheduled visits, treatment plan, laboratory tests, and other study procedures.
3. Male and female subjects of childbearing potential and at risk for pregnancy must agree to use of a highly effective method of contraception throughout the study and for at least the 28 days after the last dose of assigned treatment.
4. Female subjects of non-childbearing potential must meet at least one of the following criteria:
a. Have undergone a documented hysterectomy and/or bilateral oophorectomy.
b. Have medically confirmed ovarian failure.
c. Achieved postmenopausal status, defined as follows: cessation of regular menses for at least 12 consecutive months with no alternative pathological or physiological cause; status may be confirmed by having a serum follicle-stimulating hormone (FSH) level within the laboratory’s reference range for postmenopausal women.
All other female subjects (including females with tubal ligations and females that do
NOT have a documented hysterectomy, bilateral oophorectomy and/or ovarian failure) will be considered to be of childbearing potential.
5. Healthy male and/or female subjects between the ages of 18 and 60 years, inclusive.
Healthy is defined as no clinically relevant abnormalities identified by a detailed medical history, full physical examination, including blood pressure and pulse rate measurement, 12-lead ECG and clinical laboratory tests.
6. Body Mass Index (BMI) of 17.5 to 32 kg/m2; and a total body weight _50 kg (110 lbs).
7. Subjects who are right handed.
8. Subjects who can read, speak, and comprehend English.
9. Subjects must be outpatients with a primary diagnosis of PTSD (DSM-V criteria 309.81). Symptom severity must be such that subjects have a Clinician-administered CAPS (_35) and CGI-S score (_3) at both Screening and Day 1 (baseline) visits to qualify for clinical trial entry.
PTSD must be the primary psychiatric disorder present (confirmed by Structured Clinical Interview for DSM IV research version patient edition (SCID-RV/P) and the condition that motivated the subject to seek treatment.

| Aim of Study | We are looking for depressed participants suffering from Major Depressive disorder (MDD) or Bipolar disorder (BD) who have not improved after medication treatments for a study with an investigational novel medication aiming to relieve symptoms of depression. Participants would have visits in our clinic. Eligible participants will receive treatment at no cost and compensation for their time.

• Have a history of 3 or more failed medication treatments
• Are 21-70 years of age
• Subjects with major depression (MDD) should be willing to taper off any psychiatric medications if currently being taken

INTERNATIONAL STUDY TO PREDICT OPTIMIZED TREATMENT in ATTENTION-DEFICIT AND HYPERACTIVITY DISORDER
| Aim of Study | This is a multi-center, open-label effectiveness trial to identify objective indicators of treatment response in ADHD subjects (versus healthy controls) using cognitive and brain functions, brain structure and genetic measures in subjects diagnosed with ADHD.

What is PET?
PET (Positron Emission Tomography) is a non-invasive diagnostic scanning technique that provides researchers and clinicians with visual images of organ function. PET scans can detect biochemical changes in body tissues before structural damage occurs from disease. This information allows clinicians to be proactive in their treatments and enables researchers to detect early biomarkers of disease that can aid diagnosis and advance drug development. We are currently utilizing PET to study the serotonin, norepinephrine, and cannabinoid neurotransmitter systems in the following study populations:
Healthy Controls: male or female subjects between 18-55 years of age, without a history of any type of neurological illness or injury or any current or prior mental health or substance use disorder.
PTSD: male or female subjects, ages 18-55, who are suffering from posttraumatic stress disorder (PTSD). Individuals who have had reactions to major traumatic life events are also encouraged to participate.
Depression: male or female subjects, ages 18-55, who are suffering from depression.
Alcohol Dependence: male or female subjects between 18-50 years of age, suffering from alcohol dependence or in partial remission. Subjects may participate in an inpatient or outpatient setting, but must be abstinent from alcohol for 4-6 weeks prior to MR and PET scanning.
| Aim of Study | Aim of the studies: to examine the association between disorders such as PTSD, depression, and alcohol dependence, and brain function using MRI and PET.

Yale University, New Haven, CT and VA Medical Center, West Haven, CT | Inclusion Criteria | We are looking for male or female subjects, ages 18-55, who are healthy or suffering from depression, PTSD, or alcohol dependence. In addition, participants must have a willingness to participate in imaging studies including 1 PET scan and 1 MRI scan.

INTERNATIONAL STUDY TO PREDICT OPTIMIZED TREATMENT in ATTENTION-DEFICIT AND HYPERACTIVITY DISORDER
Is your child experiencing symptoms of ADHD? Problems paying attention, hyperactivity or impulsivity?
| Aim of Study | This is a multi-center, open-label effectiveness trial to identify objective indicators of treatment response in ADHD subjects (versus healthy controls) using cognitive and brain functions, brain structure and genetic measures in subjects diagnosed with ADHD.
| Inclusion Criteria | To be eligible for screening children must:
Be 6-17 years of age
Provide informed consent from a parent and/or legally recognized guardian
Meet the DSM IV criteria for ADHD
Be a candidate for treatment with methylphenidate

| Aim of Study | Mystery Shopping
Many businesses hire mystery shoppers to see how well they are serving the customers. A mystery shopper is someone who is paid to visit an establishment such as a retail store, bank, or restaurant, for the purpose of measuring the customer experience. Most mystery shoppers sign up with a company that coordinates the assignments. When you are given an assignment as a mystery shopper, you will be given details on conducting your visit. During your visit, you may be required to purchase an item (meal at a restaurant, movie tickets, etc.) You will be reimbursed for your purchase. After your visit, you will complete a report detailing your experience. If you properly complete your assignment, you will be paid a fee for your efforts.
